The main problem aside of safety concerns is work - it requires a lot of resources to launch one Shuttle.

It would be possible, but I think they don't do it because of safety.
Not possible! There's only one qualified Launch Team and that team is more than busy enough with one stack.

From start tanking to T0 it takes about 9½ hours. But the Launch Team members actually report for duty about 3 hours prior to that, leading to about 12½ hours of actual on-duty time for them.

This is usually why they never go for a "three in a row" launch attempts, management want to give the team some time off to rest.

There are other teams such as vehicle securing and scrub turnaround teams that takes over from the Launch Team in the event of a scrub to give the Launch Team some time off, but these teams are never on duty for longer than 8 hours.

This is just some of the personnel problems, there are other technical problems as well that prevents this.
